    Brief

    NIBRT wish to procure an automated solution for the optimisation of downstream process development. The ideal solution would comprise of robotic liquid handling and scaled down chromatography capability to enable optimisation of purification parameters using design of experiments and subsequent seamless scale up to lab or pilot scale chromatographic purification. The automated platform will be used for optimisation of downstream processing of recombinant proteins, monoclonal antibodies, viral vectors and other biopharmaceutical formats. Ideally, the robotic capability of the platform should also be capable of automated sample preparation for analysis of the samples generated from on deck downstream process optimisation.

Roboten skal primært brukes til konstruksjon av NGS-biblioteker og klargjøring av disse til sekvensering, inkludert overføring av DNA-templat, tilsetting av mastermikser og primere, rensing av PCR-produkter, automatisert kvantifisering av DNA, individuell normalisering av biblioteker og sammenslåing av biblioteker. Instrumentet må ha høy kapasitet, pipetteringspresisjon og -nøyaktighet, selv ved lave reaksjonsvolumer, og et intuitivt brukergrensesnitt. Det er også viktig at instrumentdesignet tillater effektiv rengjøring og grundig dekontaminering, da det skal brukes post-PCR. Planlagt gjennomstrømning er 20 000–30 000 prøver per år, hovedsakelig i 4 x 96-brønnsplater, med fremtidig mulighet for 384-brønnsplater. Tildelingskriteriene er kostnad (40%), kvalitet (30%) og klima- og miljøhensyn (30%). Kvalifikasjonskrav inkluderer referanser på relevant erfaring fra oppdrag eller leveranser av automatisert væskehåndtering fra de siste 3–5 årene.
